Liver Physiol. 37): G614–G617, 1998.—The humoral arm of the mucosal immune system is principally composed of locally synthesized polymeric IgA, whose Fc portion is adapted for binding to the polymeric immunoglobulin receptor that is expressed on the basolateral surface of mucosal epithelial cells, including enterocytes. This receptor mediates the endocytosis and transcytosis of polymeric IgA, which allows IgA to function in host defense at three anatomic levels in relation to mucosal epithelium: IgA antibodies in the lamina propria can bind antigens and excrete them through the epithelium into the lumen; antiviral IgA antibodies in transit through epithelial cells can inhibit virus production by an intracellular action; and IgA antibodies secreted into the lumen can prevent antigens and microbes from adhering to and penetrating the epithelium. The ways in which IgA antibodies function in mucous membranes provide challenging investigative opportunities for cell physiologists and cell biologists.

متن کاملReceptor-mediated Immunoglobulin G Transport Across Mucosal Barriers in Adult Life: Functional Expression of FcRn in the Mammalian Lung
Mucosal secretions of the human gastrointestinal, respiratory, and genital tracts contain the immunoglobulins (Ig)G and secretory IgA (sIgA) that function together in host defense. Exactly how IgG crosses epithelial barriers to function in mucosal immunity remains unknown. Here, we test the idea that the MHC class I–related Fc-receptor, FcRn, transports IgG across the mucosal surface of the hum...

متن کاملClearance of Citrobacter rodentium requires B cells but not secretory immunoglobulin A (IgA) or IgM antibodies.
Citrobacter rodentium, a murine model pathogen for human enteropathogenic Escherichia coli, predominantly colonizes the lumen and mucosal surface of the colon and cecum and causes crypt hyperplasia and mucosal inflammation. Mice infected with C. rodentium develop a secretory immunoglobulin A (IgA) response, but the role of B cells or secretory antibodies in host defense is unknown. To address t...
متن کاملCutting edge: lung mucosal Th17-mediated responses induce polymeric Ig receptor expression by the airway epithelium and elevate secretory IgA levels.
Polymeric Ig receptor (pIgR) is a central player in mucosal immunity that mediates the delivery of polymeric IgA and IgM to the apical surface of epithelial cells via transcytosis. Emerging evidence suggests that Th17 cells not only mediate autoimmunity but also play key roles in mucosal host defense against pathogens. We demonstrate that OVA-specific CD4(+) Th17 cells, in addition to causing n...
